Functional differences between primary monocyte-derived and THP-1 macrophages and their response to LCPUFAs

In immune cell models, macrophages are one of the most frequently used types. THP-1 cells often as model to study macrophage function, however they may act differently from primary human monocyte derived (MDMs). this study, we investigated intrinsic baseline differences between line and MDMs. Additionally, studied difference in response treatment with long-chain polyunsaturated fatty acids (LCPUFAs): well-described immunomodulators. Although amount that phagocytose were similar types, MDMs consumed significantly more E. coli bioparticles compared macrophages. M1 macrophages, IL-12 secretion was almost fifty times higher by thereby increasing IL-12/IL-10 ratio. Despite this, IL-10, showing it is still a suitable type. Cytokine profiles differed M2 LCPUFAs, alike. Interestingly, secreted less IL-10 CCL22 when treated whereas LCPUFAs showed no secretion. conclusion, an setting, both models.
